Trump took to Twitter late on Friday night to deny the claims and renew his attacks on Wolff and his former top aide Steve Bannon, who was quoted in the book.
"My support is also unwavering for the president and his agenda - as I have shown daily in my national radio broadcasts, on the pages of Breitbart News and in speeches and appearances from Tokyo and Hong Kong to Arizona and Alabama", Bannon said.
"I regret that my delay in responding to the inaccurate reporting regarding Don Jr has diverted attention from the President's historical accomplishments in the first year of his presidency", Mr Bannon said Monday.
"The salacious new book was written by a reporter for the left-wing Guardian, whom Bannon saw fit to entrust with extraordinary access to the Trump White House".
"The one description that everyone gave, everyone has in common - they all say he is like a child", Wolff said.
"Only because I went to the best colleges, or college".
Trump, answering questions from reporters at Camp David later, called Wolff a "fraud" and said the book is "a complete work of fiction".
Bannon said Sunday he is behind Trump 100 percent.
Wolff also hit back at claims the book is untruthful.

Trump, in a series of morning posts on Twitter, said his Democratic critics and the USA news media were bringing up the "old Ronald Reagan playbook and screaming mental stability and intelligence" since they had not been able to bring him down in other ways.
